Latest photoacoustic imaging technology provides a complete cross-sectional image of small animal organs

Many small experimental animals, most commonly mice, are indispensable for new drug development, physiology, pathology, and preclinical research. Researchers from Duke University and the University of Washington, St. Louis have developed a technique called single-pulse photoacoustic computed tomography (SIP-PACT). This is a new combination of photoacoustic technology that delivers amazingly high resolution for real-time tomography of live small animals. The new technology can observe the appearance of the living organ, the blood is flowing, the nerve is discharging, and the tumor is growing. The study was published in Nature Biomedical Engineering.

Previously, whole body imaging of these animal models relied on classical non-optical methods including, for example, magnetic resonance imaging (MRI), X-ray computed tomography (X-ray CT), positron emission tomography (PET) or single photon emission computed tomography. Photography (SPECT) and ultrasound tomography (UST). While these techniques can provide deep penetration, they have various limitations. In the case of MRI, achieving microscopic resolution requires expensive high magnetic fields and long data acquisition times ranging from a few seconds to a few minutes, which is too slow for kinetic studies. X-ray CT lacks functional contrast. PET and SPECT have poor spatial resolution when used alone. In addition, X-ray CT, PET and SPECT deliver strong ionizing radiation, which is not suitable for whole body use in vivo.

The photoacoustic imaging technology we introduced today breaks the long-standing obstacles to the resolution and speed of whole body imaging of small animals. The new technology quickly provides a complete cross-sectional image of the internal organs of small animals.

Traditional optical microscopes can display fast, high-resolution images of tissue absorption and emission, but light penetration through tissue is only a few millimeters, severely limiting its application. Ultrasound can penetrate deep into the tissue, but it cannot read the chemical composition and light information of the tissue. SIP-PACT technology integrates photoacoustic imaging into a platform that uses very short laser pulses to enter the tissue, which generates ultrasound due to tissue heating and expansion, which the ultrasound transducer can then detect to render the image.

This is a safe imaging technology that currently achieves depths of up to 5 cm and sub-millimeter resolution while retaining the functionality of an optical microscope. The upgraded device can cross-sectionally scan adult rats at a frequency of 50 times per second and image the details of the internal workings of the living body at a resolution of 120 microns.
